A Humoral Immune Response Alters the Distribution of Enzyme Replacement Therapy in Murine Mucopolysaccharidosis Type I

Abstract: Antibodies against recombinant proteins can significantly reduce their effectiveness in unanticipated ways. We evaluated the humoral response of mice with the lysosomal storage disease mucopolysaccharidosis type I treated with weekly intravenous recombinant human alpha-l-iduronidase (rhIDU). Unlike patients, the majority of whom develop antibodies to recombinant human alpha-l-iduronidase, only approximately half of the treated mice developed antibodies against recombinant human alpha-l-iduronidase and levels w… Show more

“…In long-term administration trials with RTB-enzyme fusions, anti-RTB and anti-cargo enzyme immunoglobulin levels in terminal serum have been measured by ELISA. Consistent with data for infusions with recombinant mammalian-cell-derived enzyme [66][67][68], elevated levels of total IgGs reacting against the cargo were detected in the respective KO mice treated with enzyme:RTB. In contrast, ELISA using recombinant RTB as a capture molecule shows no antibody response against the RTB lectin carrier [63,69].…”

“…The poor and inconsistent anti-IDUA humoral response described in MPS-I mice was previously demonstrated, and it was independent of the amount of injected enzyme: high doses of enzyme also failed in promoting a homogeneous immunity in treated mice. 24 Moreover, the percentage of Ab-positive mice was shown to be higher when ERT was applied in adult compared to newborn MPS-I mice. 25 However, the ex vivo HSC GT required the treatment of mice at 4 weeks of age, thus limiting the use of ERT as a model for dissecting the role of pre-existing anti-IDUA immunity on GT.…”

“…With continued nurturing and support, she has now obtained multiple grants and published several manuscripts, and has taken a compound to clinical trials. [1][2][3][4] • A senior, highly successful basic scientist requested a consultation to discuss her application when it was not awarded upon her first submission. Following the consultation and being steered toward a translational application for her project, she was awarded on a subsequent submission.…”
