A host-mediated in vivo/in vitro assay with peritoneal murine macrophages for the detection of carcinogenic chemicals

Abstract: We have developed a host-mediated assay system for the detection of the transforming action of chemical carcinogens on peritoneal macrophages. Directly as well as indirectly acting carcinogenic substances administered intraperitoneally to NMRI mice could be examined in this way. Resident macrophages were recovered by peritoneal lavage from treated and untreated mice and were cultured in soft agar. After 5-6 days normal and transformed cells could be distinguished. Statistical analysis comparing cells, for exam… Show more

“…Exposure to chemicals, such as alpha-naphthylamine and N-methyl-N-nitrosourea, can render macrophages themselves tumorigenic. Indeed, peritoneal murine macrophages exposed to these chemicals can give rise to immortal cell lines with tumorigenic activity in athymic nu/nu mice (112). But other agents, besides chemical ones, can also promote the tumorigenic activity of macrophages.…”

“…Normal peritoneal macrophages were isolated from NMRI-mice after intraperitoneal injection of LPS [13]. Both cell types were maintained in DMEM containing 10% fetal calf serum (FCS) and 1% stock solution of penicillin/streptomycin in a humidified incubator at 37°C and an atmosphere of 5% CO,.…”

“…This review focuses on a host-mediated in vivo/in vitro model assay system developed by our group (9). A major advantage of host-mediated models compared to solely in vitro studies is that they allow researchers to monitor the biotransformation of chemical mutagens through metabolic activation or detoxification and elimination.…”

“…In this review, the ability of our in vivo/in vitro assay system (9) to detect the transforming potential of chemicals was compared to that of established genotoxicity and mutagenicity test systems, such as the Ames test ( 6), micronucleus assay (10) and Comet assay (7,11). Additionally, a comparison of literature carcinogenicity data and current classifications by the IARC with respect to carcinogenic potential was made.…”
