A Highly Selective Fluorescent Probe for Quantitative Detection of Hydrogen Sulfide

Abstract: We designed and synthesized a highly selective and sensitive fluorescent probe for hydrogen sulfide (H2S) based on the one step nucleophilic addition reaction between H2S and electron-poor C=C double bond. Our proposed probe displayed high selectivity for H2S over other analytes including cysteine (Cys) and glutathione (GSH), which might be because the steric hindrance of H2S is less than other thiols. Additionally, a linear relationship between fluorescence intensity and the concentrations of Na2S (0 -450 μM)… Show more

“…In addition to this modified colorimetric technique, we also developed a reverse-phase HPLC (RP-HPLC) method for quantifying H 2 S in aqueous solution. There are many methods in the literature employing HPLC for quantification of H 2 S in tissues and plasma [ 61–63 ]. These methods use monobromobimane reagent, which requires long and tedious preparation steps with long retention times [ 64 ].…”

“…Compound 1 was synthesized according to previous methods (Ding et al, 2013 ). 9-Anthracenecarboxaldehyde (82.4 mg, 0.4 mmol) and acetone (35 mg, 0.6 mmol) were dissolved in ethanol (50 mL).…”

“…Using this approach, Ding and Zhu reported a benzimidazole conjugated to an electron poor α,β-unsaturated ketone to generate 687 (Figure ). This probe reacted with H 2 S to abolish fluorescence quenching from the conjugated nitrobenzene motifs and provide a fluorescence enhancement at 450 nm with good selectivity for H 2 S over competing thiols. Shang used this same approach to develop anthracene-based 688 , which was investigated in DMSO .…”
