A high-resolution gene expression map of theArabidopsisshoot meristem stem cell niche

Abstract: The shoot apical meristem (SAM) acts as a reservoir for stem cells. The central zone (CZ) harbors stem cells. The stem cell progenitors differentiate in the adjacent peripheral zone and in the rib meristem located just beneath the CZ. The SAM is further divided into distinct clonal layers: the L1 epidermal, L2 sub-epidermal and L3 layers. Collectively, SAMs are complex structures that consist of cells of different clonal origins that are organized into functional domains. By employing fluorescence-activated ce… Show more

“…Most studies have been focused on the cross-talk between phytohormones, such as auxin and cytokinin, and gene expression in these processes (25,26). Little is known whether, and if so how, external signals like light and temperature contribute to meristem regulation.…”

“…Furthermore, KAN proteins are excluded from the P0, which is defined by high levels of the KAN1-repressed target PIN1, and instead accumulate in a ring below initiating organs (Merelo et al, 2013;Yadav et al, 2014).…”

“…The exclusion of KAN proteins from the P0 allows the nonpolar accumulation of AS1-AS2 and the initial nucleation of PRC2-mediated H3K27me3 at the promoter of MIR166A on both sides of the primordium. As the primordium develops, KAN expression extends into its abaxial side (Yadav et al, 2014), directly repressing AS2 expression and restricting further deposition of H3K27me3 at MIR166A solely to the adaxial side. This polarizes the production of mature miR166 and consequently limits HD-ZIPIII accumulation to the adaxial side.…”

“…52,46 Among those are ANT, CLF, AS1, AS2 that are expressed mainly in the growing organ and, CUC1, 2 and 3, the expression of which is restricted to the boundary domain. 43,46,52,54 The pattern of stress anisotropy is represented by color bar with green corresponding to the regions with lower stress anisotropy (central zone and organ) and orange in the boundary domain where stress anisotropy is maximal. This stress and gene expression pattern correlates with the distribution of the plant hormone auxin that has its local maximum where new organ initiates and its local minimum at the organ boundary.…”
