A high performance liquid chromatography method for simultaneous detection of 20 bioactive components in tea extracts

Abstract: A high performance liquid chromatography method for simultaneous detection of 20 bioactive components in tea extractsTea is the second most widely consumed beverage and contains various bioactive compounds. A simple method to analyze these compounds is of great scientific and commercial interest. In this work, a 30 min HPLC method was developed using a simple gradient elution system, and the mobile phases and elution gradients were optimized. This method separated 17 polyphenols and three alkaloid compounds in… Show more

“…The chemical compounds were separated with an Agilent C18 chromatographic column (250 × 4.6 mm, 5 µm) protected with a Phenomenex C18 guard column (10 mm × 4.6 mm, 5 µm; Phenomenex, Torrance, CA, USA) by reference to the chromatographic conditions described in the studies of Nian et al. (2019) and Guo et al. (2018).…”

“…An Agilent 1200 HPLC system (Agilent Technologies, Santa Clara, CA, USA) consisted of infinity binary pump, integrated vacuum degasser, autosampler, thermostated column compartment, and diode array detector (DAD) were used to determine the contents of caffeine, theobromine, GA, and the catechins, including C, EC, EGC, GC, ECG, GCG and EGCG, as well as the flavonoids including quercetin, kaempferol, myricetin and rutin. The chemical compounds were separated with an Agilent C18 chromatographic column (250 × 4.6 mm, 5 µm) protected with a Phenomenex C18 guard column (10 mm × 4.6 mm, 5 µm; Phenomenex, Torrance, CA, USA) by reference to the chromatographic conditions described in the studies of Nian et al (2019) and Guo et al (2018).…”

“…The concentrations of the 2 alkaloids and 16 polyphenols in tea leaves were determined using an Agilent 1200 series HPLC system having a Poroshell 120 EC-C18 column (4.6 × 100 mm, 2.7 μm) fitted with a C18 guard column (Agilent Technologies, Santa Clara, CA, USA). The detailed elution conditions have been described previously ( 19 ). The contents of 18 amino acids in tea leaves were measured in accordance with previous reports ( 20 , 21 ).…”

“…The extraction was performed in a water bath (at 85℃) for 90 min. The extractions were diluted to 50 ml and filtered through a 0.2‐μm nylon filter and then analyzed directly by HPLC as described in our previous work (Nian et al., 2019 ).…”
