A High-Content Screen for the Identification of Plant Extracts with Insulin Secretion-Modulating Activity

Abstract: Bioactive plant compounds and extracts are of special interest for the development of pharmaceuticals. Here, we describe the screening of more than 1100 aqueous plant extracts and synthetic reference compounds for their ability to stimulate or inhibit insulin secretion. To quantify insulin secretion in living MIN6 β cells, an insulin–Gaussia luciferase (Ins-GLuc) biosensor was used. Positive hits included extracts from Quillaja saponaria, Anagallis arvensis, Sapindus mukorossi, Gleditsia sinensis and Albizia j… Show more

“…After being released from cells, insulin can be measured via radioimmunoassay [85,86] or ELISA. Recently, a luminescent alternative to detect insulin was described by Hager et al and Kalwat et al [87,88]. Despite continuous improvements in measuring glucose and glucose-associated processes, a relevant challenge is to fully understand the physiological and pathological roles of blood glucose levels and their impact on health conditions and disease.…”

“…Insulin secretion was measured by luminescence as described. [37] GLUT4 Cell Surface Localization: GLUT4 cell surface localization was measured by total internal reflection fluorescence (TIRF) microscopy allowing for selective excitation of membrane-adjacent fluorescently tagged GLUT4 as previously described. [38] Briefly, HeLa GLUT4-myc-GFP cells were grown in RPMI 1640 in 96-well imaging plates (4 × 10 4 cells per well) overnight.…”

“…Secreted GLuc activity in this manner has been well-established as a proxy for insulin release [19,[25][26][27][28][29]. Our approach involved treating InsGLuc-MIN6 beta cells for 24 h with compounds followed by a brief wash-out prior to testing their sustained impact on insulin secretion.…”
