Borrelia burgdorferi, the agent of Lyme borreliosis, is a spirochetes transmitted by ticks to humans and animals. Its cultivation in vitro in tick cells allows studies of its biology and provides methodology for future research in Brazil, and for the isolation of Borrelia spp. We examined in vitro the characteristics of embryonic cells of Rhipicephalus microplus and Amblyomma cajennense in cell culture and investigated the suitability of embryonic cells as a substrate for cultivation of B. burgdorferi. Subcultures were prepared from primary cultures of embrionary cells of R. microplus and A. cajennense maintained in Leibovitz's (L-15) complete medium at 28 °C and 31 °C, respectively. When a monolayer had formed, the L-15 was replaced with Barbour-Stoener-Kelly medium for experiments to infect cell cultures with B. burgdorferi. After 72 hours of cultivation, the spirochetes were counted using an inverted phase contrast microscope and dark-field illumination (400×). Survival, multiplication and the adherence of B. burgdorferi for embryonic cells of R. microplus and A. cajennense were observed. B. burgdorferi cultured with embryonic cells of R. microplus grew on average to a density (final count) of 2.4 × 10 7 spirochetes/mL, whereas in cell-free culture, an average of 2.5 × 10 7 spirochetes/mL were counted. When cultivated with A. cajennense cells, the final count of spirochetes was on average 1.7 × 10 7 spirochetes/mL, while spirochetes cultured under cell-free conditions replicated on average of 2.

ResumoA maioria das atividades de treinamento militar transcorre em ambiente de risco para aquisição de doenças de transmissão vetorial, incluindo importantes zoonoses e doenças emergentes/reemergentes. O objetivo deste estudo foi relatar a ocorrência de anticorpos homólogos da classe IgG anti-Borrelia burgdorferi em equinos de uso militar no Brasil, assim como analisar os fatores de risco associados à soropositividade nos animais. Foram estudados dois plantéis de equinos, sendo um no município de Resende, estado do Rio de Janeiro, tendo sido encontrada prevalência de 29,9%, e outro no município de São Borja, estado do Rio Grande do Sul, com prevalência de 44,7%, totalizando 474 animais e prevalência de 39,24%. Na análise dos fatores de risco associados, observou-se que a infestação por carrapatos constituiu-se no fator de risco para a soropositividade. A evidência de circulação de Borrelia spp. com soroprevalência nos equinos reforça seu papel como sentinela, sendo fator de alerta para potencial ocorrência de casos humanos. Os resultados obtidos reforçam a importância da vigilância epidemiológica dos carrapatos vetores e sua associação tanto com agentes infecciosos de risco para a saúde do efetivo militar humano, como nos equinos de uso militar nas regiões avaliadas. Isto também realça a necessidade da adoção de medidas de proteção coletivas e individuais visando mitigar o risco de doenças transmitidas por carrapatos em atividades militares. AbstractMost military training activities take place at risk for the acquisition of vector-borne diseases, including important zoonosis and emerging/reemerging diseases This study aimed to analyze the seroprevalence of homologous antibodies antiBorrelia burgdorferi in horses for military use in Brazil, as well as to analyze the risk factors associated with seropositivity in animals. Two herds of horses for military use were studied, one in the city of Resende, state of Rio de Janeiro, having been found prevalence of 29.9%, and in São Borja, state of Rio Grande do Sul, with prevalence 44,7% to 474 animals and prevalence of 39,3%. In the analysis of risk factors, it was observed that the tick infestation constituted risk factors for seropositivity. Evidence of Borrelia spp. circulation with seroprevalence in horses reinforces its role as sentinels, and alert factor for potential occurrence of human cases. These results support the importance of epidemiological surveillance of vector ticks and its association with agents of risk to the health of horses for military use, in the evaluated areas. This also highlights the need to adopt collective and individual protection measures to mitigate the risk of tick-borne diseases in military activities.
