A golden phoenix arising from the herbal nest — A review and reflection on the study of antimalarial drug Qinghaosu

Abstract: Qinghaosu (QHS) and its derivatives are a new generation of antimalarial drugs characterized by fast action, high efficacy, and good tolerance. This feature article states the discovery of QHS from traditional Chinese medicine qinghao (Artemisia annua L.) and reviews the progress during the past four decades in the research of phytochemistry of A. annua, chemical reactions and biotransformation of QHS, chemical synthesis and biosynthesis of QHS, synthesis and antimalarial activity of QHS derivatives and analog… Show more

“…After screening of over 5000 traditional Chinese medicines, Qinghaosu (QHS), an antimalarial principle, was isolated from Artemisia annua L in 1972 [1,2] . At the end of 1975, its unique chemical structure was elucidated, as a sesquiterpene lactone bearing a peroxy group, quite different from that of all known antimalarial drugs [3] .…”

“…QHS suppository in 1986; artemether oil injection and sodium artesunate aqueous injection in 1987 were approved as new antimalarial drugs. Since then, other new antimalarial drugs such as dihydroartemisinin, coartem (artemether and benflumetol), co-naphthoquine (naphthoquine phosphate and QHS), compound-dihydroartemisinin were successively developed in China [1,2] .…”

“…Qinghaosu (QHS, artemisinin, 1 ) is a sesquiterpene endoperoxide discovered by Chinese scientists in the early 1970s (Figure ) . Its structure is completely different from that of traditional antimalarials and shows high potency in the treatment of even multidrug resistant cases.…”

“…Many natural products have been found to possess antineoplastic activity with minimal side effects [3]. Artemisinin, a well-known anti-malarial agent, has been reported to possess a potent and broad antitumor spectrum against human cancer cell lines [4]. However, the drawback, such as drug resistance or unsatisfactory metabolism, constrains artemisinin to be employed as an antitumor drug clinically.…”
