A Global Analysis of the Receptor Tyrosine Kinase-Protein Phosphatase Interactome

Abstract: SUMMARY Receptor tyrosine kinases (RTKs) and protein phosphatases comprise protein families that play crucial roles in cell signaling. We used two protein-protein interaction (PPI) approaches, the Membrane Yeast Two-Hybrid (MYTH) and the Mammalian Membrane Two-Hybrid (MaMTH), to map the PPIs between human RTKs and phosphatases. The resulting RTK-phosphatase interactome reveals a considerable number of previously unidentified interactions and suggests specific roles for different phosphatase families. Additiona… Show more

“…This approach is bolstered by the fact that surveys of protein-protein interactions have revealed many interactions that, when disrupted, are deleterious. [39][40][41][42][43][44] Moreover, protein interactions can reveal defects in protein folding and stability, which helps to explain why approximately one-third of disease-related variants in proteins with multiple interaction partners disrupt all of the interactions of that protein. 45 Variants of protein-coding genes can also result in pathogenicity by altering splicing.…”

“…The study of PTPs has long suffered from a paucity of tools for probing and measuring their intracellular activities 46 . In this study, we developed a photoswitchable variant of PTP1B and used it to exert spatiotemporal control over the phosphorylation state of a genetically encoded biosensor in living cells.…”

“…docking motifs or different subunit combinations) provides substantial specificity. 158 There has been some success in examining phosphatase interaction partners with a mammalian two-hybrid system 159 or cross-linking mass spectrometry, 160 but because these studies indicate what proteins simply associate with phosphatase complexes, additional work is needed to verify their identities as substrates. Another strategy involves mutation of the phosphatase catalytic domain so that it binds tightly to potential substrates.…”
