A Genomic Signature Reflecting Fibroblast Infiltration Into Gastric Cancer Is Associated With Prognosis and Treatment Outcomes of Immune Checkpoint Inhibitors

Abstract: Background: The immunotherapy efficacy in gastric cancer (GC) is limited. Cancer-associated fibroblasts (CAFs) induce primary resistance to immunotherapy. However, CAF infiltration in tumors is difficult to evaluate due to the lack of validated and standardized quantified methods. This study aimed to investigate the impact of infiltrating CAFs alternatively using fibroblast-associated mutation scoring (FAMscore). Methods: In a GC cohort from Affiliated Hospital of Jiangsu University (AHJU), whole exon sequencin… Show more

“…In an additional AHJU GC cohort with transcriptome data from 34 patients [ 18 , 19 ], DEGs were identified between the high and low subgroups (median value as the cutoff) of CHAF1A expression, based on the criteria of adjusted p -value < 0.05, and log 2 (fold change) > 1 (Fig. 4 A, B).…”

“…Transcriptome data from 34 patients in an additional AHJU GC cohort were used to explore the signaling network associated with the object gene [ 18 , 19 ]. Transcriptome data were stored in the European Genome-Phenome Archive ( https://ega-archive.org/ ), with the identification number EGAD00001004164.…”

“…CAFs are the protagonists of TME in gastric cancer, associated with chemoresistance and dismal prognosis. Therefore, CAFs hold great promise as a potential therapeutic target in treating gastric cancer ( Liu et al, 2021a ; Lu et al, 2022 ). Despite the promise of CAF-targeting strategies in cancer, the lack of a complete understanding of the CAFs and their interaction with cancer cells may pose challenges in drug discovery.…”

“…Data from other cohorts have been previously published and were acquired and preprocessed as described elsewhere. 19 , 20 Immune-associated indices, including tumor mutation burden (TMB), tumor neoantigen burden (TNB), microsatellite instability (MSI), and immune subtype, have been previously defined and determined. 19 , 20 , 23 …”
