A Genomewide Screen for Tolerance to Cationic Drugs Reveals Genes Important for Potassium Homeostasis in Saccharomyces cerevisiae

Abstract: Potassium homeostasis is crucial for living cells. In the yeast Saccharomyces cerevisiae, the uptake of potassium is driven by the electrochemical gradient generated by the Pma1 H ؉ -ATPase, and this process represents a major consumer of the gradient. We considered that any mutation resulting in an alteration of the electrochemical gradient could give rise to anomalous sensitivity to any cationic drug independently of its toxicity mechanism. Here, we describe a genomewide screen for mutants that present alter… Show more

“…An altered K þ homeostasis was shown to affect the sensitivity to cationic drugs 28 and thus could be also related to the Cs þ phenotype of sec22D. However, in contrast to nutrient-deficient medium, the early uptake time course of Rb þ is identical for wild type and sec22D in YPD-rich medium ( Supplementary Fig.…”

“…The mitochondrial marker succinate dehydrogenase was assayed by quantifying reduced p-iodonitrotetrazolium violet 48,51 . Finally, carboxypeptidase Y and a-D-mannosidase were employed as markers for the vacuolar lumen and membrane, respectively 28,52,53 . Carboxypeptidase Y activity was based on the release of leucine from N-CBZ-L-Phe-L-Leu.…”

“…Prior studies established that trk1Δtrk2Δ yeast are sensitive to the toxic cations hygromycin B (an aminoglycoside antibiotic), spermine, and tetramethylammonium (53). This phenotype arises from increased cellular uptake of the toxins in trk1Δtrk2Δ yeast because the membrane is hyperpolarized when potassium entry is limited.…”
