A genome‐wide set of 106 microsatellite markers for the blue tit (Cyanistes caeruleus)

Abstract: We have characterized a set of 106 microsatellite markers in 26-127 individual blue tits (Cyanistes caeruleus), and assigned their location on the zebra finch (Taeniopygia guttata) and on the chicken (Gallus gallus) genome on the basis of sequence homology. Thirty-one markers are newly designed from zebra finch EST (expressed sequence tags) sequences, 22 markers were developed by others from EST sequences using different methods and the remaining 53 loci were previously designed or modified passerine markers. … Show more

“…More precisely, loci that were designed based on or showed homology to zebra finch ( Taeniopygia guttata ). Expressed sequence tags (ESTs) were considered to be putatively functional, whereas markers designed using traditional cloning methods and with no homology to avian ESTs were considered to be neutral (for more details on loci development, see Olano‐Marin et al., 2010). It must be emphasized two important questions regarding this approach.…”

“…Primer sequences and annealing temperatures for the microsatellites are given in Table S1 (see also Olano-Marin et al [29] and Hansson et al [30]). The PCR-products were separated and visualized using an ABI 3730 capillary sequencer (Applied Biosystems), and the genotypes were scored with Genemapper 4.0 (Applied Biosystems).…”

“…In blue tits, an extensive number of nuclear microsatellites have been developed [29] and a first generation linkage map has been constructed [30]. This enabled us chose a set of markers with known linkage map position for our analyses.…”

“…nlm.nih.gov/mapview/) genome assemblies, respectively (cf. Dawson et al, 2007;Backströ m et al, 2008a;Olano-Marin et al, 2009). This genotyping strategy enables comparisons of synteny, gene order and pattern of recombination in homologous chromosomal regions of Passeriformes and Galliformes, two phylogenetically highly diverged avian lineages (cf.…”
