A genome-wide comprehensively analyses of long noncoding RNA profiling and metastasis associated lncRNAs in renal cell carcinoma

Abstract: Recently, a growing number of studies have indicated that long noncoding RNAs (lncRNAs) are emerging as new critical regulators of tumorigenesis and prognostic markers in multiple cancers. However, the expression pattern of lncRNAs and their contributions in renal cell carcinoma (RCC) remains poorly understood. In this study, we performed a genome-wide comprehensively analysis of lncRNAs profiling and clinical relevance to provide valuable lncRNA candidates for the further study in RCC. RCC and non-tumor tissu… Show more

“…Among these increased lncRNAs, we found that DUXAP8 and LINC00460 are consistently upregulated in TCGA data and three GEO datasets (Figure 4A,B). Interestingly, DUXAP8 has also been reported to be overexpressed in human non small cell lung cancer,24 gastric cancer25 and renal cell carcinoma,26 while LINC00460 expression is upregulated in nasopharyngeal carcinoma27 and promotes carcinogenesis in ESCC 28. Therefore, we chose these two lncRNAs for further analyses and validation.…”

“…For instance, the pseudogene DUXAP8 promotes non-small-cell lung cancer cell proliferation, and invasion by epigenetically silencing early growth response 1 and Ras homolog family member B (11). Pseudogene DUXAP8 was significantly associated with overall survival time of patients with renal cell carcinoma (RCC), and the knockdown of DUXAP8 may impair RCC cells invasive ability in vitro (17). Increased DUXAP8 promoted cell proliferation and invasion through epigenetically silencing pleckstrin homology domain contain O1 expression by binding with enhancer of zeste 2 polycomb repressive complex 2 subunit and SUZ12, polycomb repressive complex 2 subunit in gastric cancer cells (12).…”

“…Accumulating evidence has demonstrated that abnormal expression of lncRNAs plays an important role in the development and progression of tumours [30,31]. LncRNAs have become a hot topic as diagnostic markers of diseases.…”
