“…This result confirmed that RST1 functions as a suppressor of S-PTGS comparable with other proteins involved in RNA degradation 25,5557,59,63 . Of note, the role of RST1 as S-PTGS suppressor is also supported by an independent study published during the reviewing process of our paper 64 .…”

“…To test the genetic requirements for the semi-lethal effects of MHAL overexpression in an unbiased manner, we mutagenized MHAL overexpressors in the Col-0 background with ethyl methanesulfonate. When we sequenced the genomes of 72 M3 suppressor of MHAL ( somhal ) lines, we found multiple mutations in RST1 and mutations XRN4 , two loci that prevent transgene silencing (Gazzani et al, 2004; Lange et al, 2019; Li et al, 2019) (Table 3) . The most often mutated locus was ACD6 , in 37 of the 72 suppressor lines, with 28 distinct variants (Figure 3D , Table 3) .…”

“…Shown are candidate mutations, which are all G>A (C>T) changes, as typical for EMS, from 52 lines in three genes, XRN4, RST1 and ACD6, with multiple hits. Mutations in RST1 and XRN4 are known for transgene silencing (Gazzani et al, 2004;Lange et al, 2019;Li et al, 2019). In the remaining 20 lines, mutations were found, but without any overlapping genes.…”

“…RST1 is found throughout the multicellular eukaryotic organism, suggesting that RST1 inhibits conserved trafficking components rather than plant-specific factors, and likely a similar molecular breaks function of RST1 is applicable to other multicellular eukaryotic organisms. Recently, two other studies reported that RST1 also plays roles in RNA surveillance pathway, demonstrating that RST1 is a multifunctional protein in plants (Lange et al, 2019;Li et al, 2019b).…”
