A genetic analysis identifies a haplotype at adiponectin locus: Association with obesity and type 2 diabetes

Abstract: Adiponectin is a prime determinant of the status of insulin resistance. Association studies between adiponectin (ADIPOQ) gene single nucleotide polymorphisms (Snps) and metabolic diseases have been reported earlier. However, results are ambiguous due to apparent contradictions. Hence, we investigated (1) the association between ADIPOQ Snps: −11377C/G, +10211T/G, +45T/G and +276G/T for the risk towards type 2 diabetes (T2D) and, (2) genotype-phenotype association of these SNPs with various biochemical parameter… Show more

“…49 Singlenucleotide polymorphisms (SNPs) in the promoter region of the adiponectin gene may be associated with the development of insulin resistance, obesity, and T2DM. 50,51 Researchers have found H 2 S-reduced insulin sensitivity in adipocytes, and CSE inhibitors might reverse this effect. A high level of glucose (20 mmol/L) inhibited the H 2 S production of adipocytes in a time-and concentrationdependent manner.…”

“…The most significant, although without reaching p < 0.05, was rs1501299, which had already been previously analyzed in our initial list of the four pre-selected candidate SNPs in the ADIPOQ gene, therefore being consistent with our previous findings. The association between the candidate SNPs +276G > T (rs1501299) and +45T > G (rs2241766) with plasma adiponectin has been heterogeneous across diverse populations [ 29 , 41 , 46 , 47 , 48 , 49 , 50 , 95 , 96 ]. Of the four candidate SNPs analyzed, we only observed strong and additive associations for the −11391 G/A (rs17300539) polymorphism, in agreement with several studies showing significant associations for this variant [ 49 , 50 ].…”

“…Адипонектин является основной детерминантой развития резистентности к инсулину. Снижение уровня адипонектина на фоне ожирения и генетических полиморфизмов гена адипонектина (ADIPOQ) влечет за собой изменение метаболического профиля, создавая риск развития СД 2 [32]. В работе А.Ф.…”
