A general enhancement scheme in heteronuclear multidimensional NMR employing pulsed field gradients

Abstract: General pulse sequence elements that achieve sensitivity-enhanced coherence transfer from a heteronucleus to protons of arbitrary multiplicity are introduced. The building blocks are derived from the sensitivity-enhancement scheme introduced by Cavanagh et al. ((1991) J. Magn. Reson., 91, 429-436), which was used in conjunction with gradient coherence selection by Kay et al. ((1992) J. Am. Chem. Soc., 114, 10663-10665), as well as from a multiple-pulse sequence effecting a heteronuclear planar coupling Hamilto… Show more

“…Phasesensitive HSQC using Echo/Antiecho-TPPI gradient selection with decoupling during acquisition (Palmer et al, 1991;Grzesiek and Bax, 1993;Schleucher et al, 1994) was used to record the two-dimensional 1 H-15 N spectra. All HSQC spectra were acquired with 1024 ͑t 2 ͒ and 128 ͑t 1 ͒ complex points.…”

“…Footprint experiments of small RNAs on the uniformly 15 N-labeled NC protein were performed using echo-anti echo HSQC experiments [15] at 30 °C, at a concentration of 0.5 mM of each species in NMR buffer (25 mM Na-d3-acetate pH 6.5, 25 mM NaCl, 0.1 mM BME, 0.1 mM ZnCl 2 , 10 % 2 H 2 O). Each partner was prepared at a concentration of about 2 mM in NMR buffer, and the required volumes of each sample were mixed together, immediately vortexed and incubated at 30 °C for 30 min before NMR data acquisition.…”

“…2D homonuclear NOESY and TOCSY were recorded with the identical spectral width of 7800 Hz in both dimensions, using mixing times of 200 and 55 ms, respectively. A 2D 15 N^1H echo/antiecho heteronuclear single quantum correlation (HSQC) [18,19] was recorded with spectral widths of 3000 Hz and 1580 Hz in the 1 H and 15 N dimensions, respectively. 3D NOESY-HSQC [20] and 3D TOCSY-HSQC [21] were recorded with spectral widths of 4800, 2400, 8400 Hz, respectively for amide 1 H, 15 N and 1 H dimensions, and mixing times of 200 ms (NOESY) and 55 ms (TOCSY).…”
