A gender-bias-mitigated, data-driven precision medicine system to assist in the selection of biological treatments of grade 3 and 4 knee osteoarthritis: development and preliminary validation of precisionKNEE

Abstract: Introduction Osteoarthritis is a leading cause of global disability and is set to worsen with the concurrent rise in rates of obesity and an ageing population [1]. Current clinical solutions are sub-optimal with regards to their invasiveness and outcomes. Orthopaedic biologics is an emerging field that offers alternative and parallel treatment options to address this problem. Determining which patients will benefit most from these novel treatments is key in developing clinical pathways. Methods Our dataset inc… Show more

“…The use of the precisionKNEE algorithm requires external validation with an appropriate study [ 10 ]. We believe it needs to be viewed as a diagnostic tool, much like imaging, providing additional information for the clinician and the patient to navigate the complex array of treatments available.…”

“…Defining the characteristics of individuals who will respond remains a challenge. Applications and tools to support clinical decisions with predictive, data-driven, precision medicine can identify patients who will respond with reduction in pain and improvement in function to novel as well as established treatments based on pre-treatment clinicodemographic data [ 9 ]. This approach is key in developing personalized, evidence-based clinical pathways.…”

“…Several biologic therapies, including the injection of microfragmented adipose tissue, have been shown to be safe and effective [5][6][7]. The outcomes of these treatments can be variable [8], with some patients showing dramatic improvements in their symptoms while others failing to respond [9].…”

“…Defining the characteristics of individual who will respond remains a challenge. Applications and tools to support clinical decisions with predictive, data-driven, precision medicine can identify patients who will respond with reduction in pain and improvement in function to novel as well as established treatments based on pre-treatment clinico-demographic data [Heidari, Parkin, Olgiati, Meloni, Fish, Noorani, Slevin, and Azamfirei, 2021b]. This approach is key in developing personalized, evidence-based clinical pathways.…”
