SUMMARY1. The rate and volume of gastric filling was estimated in conscious ferrets by measuring the amount of milk they would drink after an overnight fast. The mean volume was 94-5 + 7-5 ml. at a rate of 13-0 + 0 74 ml./min. An intragastric infusion rate of 10 ml./min to a total of 50 ml. was selected as a standard distension stimulus.2. Action potentials were recorded from single gastric afferent fibres in the cervical vagus. All but two of thirty-six afferent units were tonically active when the stomach was deflated.3. Afferent fibres arising from receptors in the antrum showed modulation in phase with spontaneously occurring antral contractions. Afferent fibres from the corpus and fundus, however, discharged at irregular rates between 0 35 and 7-5 Hz with no correlation with the intragastric pressure rises associated with the antral contractions.4. Inflation of the stomach with 50 ml. 0-9 % NaCl at a rate of 10 ml./min stimulated antral motility and the rhythmic afferent discharge from the antrum was enhanced Receptors in the corpus and fundus increased their rate of discharge with increasing gastric volumes. Receptors in the region between the antrum and the corpus had the property of both types so that they responded to both distension and contractions.5. On distension with 0-9 % NaCl, fluid is distributed unevenly in the stomach. 80 % was accommodated in the corpus and fundus, the remainder of the fluid entering the antrum.6. The tension was measured in strips of stomach wall taken from corpus and antrum. For equal increments of stretch the development of tension was greater in the antral than in corpus strips. This physical property together with neurally mediated receptive relaxation of the corpus is the reason for the fluid distribution described above.7. It is concluded that the properties of the tension receptor are determined by their site in the stomach. Those in the body and fundus signal the degree of distension and those in the antrum signal information concerning the amplitude, rate and duration of antral contractions.

SUMMARY1. The possibility of a Na+/H+ exchange mechanism in the presence of NaF (Bond & Hunt, 1956) has been studied in acid secreting and non-acid secreting stomachs of anaesthetized cats.2. In non-acid secreting stomachs, although H+ loss and Na+ gain were both pH related, there was no constant relationship between the two.3. The H+ loss in non-acid secreting stomachs could not account for the H+ loss in histamine stimulated stomachs.4. In acid secreting stomachs the Na+ or neutral Cl-gain was only of the order of 50% of the H+ loss, and, in the presence of isosmolal NaNO3, NaF produced the same reduction in H+ but with a markedly suppressed gain in neutral chloride. 5. The NaF induced reduction in H+ output was accompanied by reductions in the total Cl-output, the K+ output and the mucosal blood flow.6. It is concluded that the reduction in H+ output from histamine stimulated stomachs is largely a result of inhibition of secretion.

In our previous papers (1, 2), we reported that a new fraction of licorice root (FM 100) showed a protective effect on ulceration in the pylorus ligated rats and gastric antisecretory activities in rats and dogs in spite of being devoid of anticholinerigc properties. FM 100 decreased non-stimulated gastric secretion volumes and acid outputs in the pylorus ligated rats and gastric acid secretion induced by instillation of peptone into the antrum separated from the fundus in acute fistula rats. Effects of histamine and methacholine on gastric secretion were partially antagonized by FM 100. On the contrary, F.100 did not exert the inhibitory action on secretion induced by crude gastrin. Then, we propounded the hypothesis that the mechanism of gastric antisecretory activity of FM 100 is probably due to inhibitory action of the release of gastrin from the antral mucosa.The aim of the present investigation was to study the mechanism of gastric antisecretory activity of F000, especially concerning the mechanism of the release of gastrin from the antral mucosa of the rat. MATERIALS AND METHODSMale Donryu rats weighing about 200 g were used for acute experiments. They were deprived of food before use but allowed free access to water overnight. Male Wistar rats weighing above 250 g were used for the preparation of chronic fistula rats . Acute fistula rats The surgical procedure was described in the previous paper (2). The gastric juice was washed out with 10 ml of saline every hour and the washing solution was titrated with 0.1 N NaOH using phenolphthalein as indicator. The acid outputs were expressed in ml of 0. 1 N HCl per hour. For stimulation of gastric secretion, intramuscular injection of gastrin like tetrapeptide (AOC-TP : t-amyloxycarbonyl Trp. Met. Asp . Phe. NH, (3) ; 500 sag/kg, i.m.), intragastric instillation of 1 ml of 20% ethanol or 1 % acetylcholine solu tion or mechanical stimulation (gastric distension induced by intragastric instillation of 10 ml of saline) were employed. Pylorus ligated rats (Shay rats)The surgical procedure of the pylorus ligated rats was identical to the method des cribed in the previous paper (2). The time of pyloric ligation was set at 4 or 5 hours . In the study of the effect of antrectomy on gastric secretion, the antral region was resected when the pylorus was ligated.
