A facility for electron microscopy of specimens in controlled environments

Abstract: A controlled environment for specimens in the electron microscope is produced within a space formed by two 20 mu m apertures 0.75 mm apart in the gap of the objective polepiece. An additional diffusion pump makes it possible to operate the microscope with gas at 1 atm pressure in the space without covering the apertures with membranes. A side entry stage permits rapid specimen changing and enables the whole of a normal grid to be scanned. Preliminary experiments using this system indicate that repeat periods l… Show more

“…Unit-cell dimensions, calibrated with a Au ° powder diffraction standard are a= 69.7 A, c= 177 A, with an orthorhombic unit cell assumed (Labaw, 1967), and are in good agreement with previous measurements of diffraction patterns from both wet and negatively stained catalase crystals of this habit (Ferrier, 1969;Wrigley, 1968;Matricardi, 1972;Unwin, 1972;Labaw, 1967;Moretz, 1973;Ward & Mitchell, 1972;Matricardi, Moretz & Parsons, 1972).…”

“…There are, for instance, studies in progress to reduce beam damage using HVEM, low-temperature specimen stages, image intensification and minimum radiation dosage devices, etc. (Williams & Fisher, 1970;Cosslett, 1971Cosslett, , 1974Ward & Mitchell, 1972;Glaeser, 1974;Hart & Yoshiyama, 1975;and others).…”

“…Swann has constructed an environmental chamber for the pole piece gap of the AEI EM7 1.0-Mev electron microscope (30) and used it extensively for metallurgical and gas reaction studies. A single-stage differentially pumped unit was constructed for the Cavendish Laboratory high-voltage microscope (36) for use in the examination of wet biological specimens.…”
