A Double-Blind Trial of Clomipramine in Premature Ejaculation

Show more

“…The current treatment choice for PE is medical treatment. Systemic treatments have included adrenergic antagonists (Beretta et al, 1986), c-aminobutyric acid, and selective serotonin reuptake inhibitors (Girgis et al, 1982;Kara et al, 1996;McMahon, 1998;McMahon and Touma, 1999 a,b;Andersson et al, 2006). Success with these agents has been variable and is associated with side effects.…”

“…Delayed ejaculation is a common side-effect of many antidepressant drugs, particularly the serotoninergic antidepressant clomipramine [ 3 ] and the SSRIs fl uoxetine, paroxetine, and sertraline [ 4 -6 ] . These drugs increase the level of synaptic serotonin, presumably inhibiting the ejaculatory refl ex, and thereby prolong intra-vaginal ejaculatory latency time (IELT).…”

“…27,28 PE has been treated with various modalities, including behavioural therapy 3 , topical anaesthetic creams or sprays, 29 and off-label use of oral pharmacotherapy, such as phosphodiesterase-5 inhibitors, 30 selective serotonin reuptake inhibitors (SSRIs) (fluoxetine, citalopram and duloxetine), 31 and a tricyclic antidepressant (clomipramine) that blocks serotonin, dopamine and norepinephrine transporters. 32 These modalities are associated with variable rates of success when taken daily, 33,34 and intracavernosal vasoactive drug injection has also been reported. 35 In contrast, some reports suggest that these drugs may be effective when received as needed.…”
