A double‐blind, placebo‐controlled trial of TRH in amyotrophic lateral sclerosis

Abstract: A double-blind, placebo-controlled trial of single doses of thyrotropin releasing hormone (TRH) was performed on 12 patients with amyotrophic lateral sclerosis. Each patient was given subcutaneous injections of TRH 150 mg or placebo, and IV infusions of TRH 500 mg or placebo at 72- to 96-hour intervals. Eight motor and functional ratings were scored at regular intervals after each injection. Side effects were seen in all patients and were obvious to patients and examiners, making true blinding impossible. Neve… Show more

“…Further, patients were divided into subgroups depending on the degree of bulbar involvement at the time of measurement. According to a bulbar involvement score [8]2 groups were differentiated, 1 with no or mild bulbar impairment (score 0–7), and the other with moderate to marked impairment (score 8–15). The disease duration (in months), the total score of the Norris scale [9], and upper motor neuron involvement [8]were correlated with gastric emptying half time (t½) and peak time (t peak ).…”

“…A muscle power rating by manual testing of the left biceps brachii muscle underlying the biopsied specimens of the skin was obtained by the following scale: 5 = normal strength; 4 = able to overcome moderate resistance; 3 = able to overcome gravity but not resistance; 2 = able to contract muscle with gravity eliminated; 1 = flicker of movement; 0 = no movement [14]. All ratings which were performed by the same examiner in individual patients were reproducible.…”
