A DNA Duplex with Extremely Enhanced Thermal Stability Based on Controlled Immobilization on Gold Nanoparticles

Abstract: The effect of DNA loadings on the thermal stability of DNA duplex immobilized on gold nanoparticles has been investigated. The modestly loaded duplexes on the gold nanoparticles showed enhanced thermal stability, as compared to that of the free duplex (without gold nanoparticles). However, the highly loaded duplex showed stability similar to that of free duplex. The stability could be controlled over a wide temperature range simply by varying the salt concentration (over 50 degrees C). Additionally, the gold n… Show more

“…42 Immobilization of DNA onto gold has also been reported to enhance its thermal stability. 48 Polyvalent and PEGylation approaches are also reported to stabilize DNA in conjunction with gold. 49,50 For example, PEG, possessing a pentaethylenehexamine at one end of each polymer chain (N6-PEG), acts as a surface-modifier in cooperation with the cations of salts in physiological conditions to reinforce the formation of the GNPs.…”

“…The unique properties, such as distance dependent surface plasmon effects that can result in dramatic color changes with extinction coefficients that are several hundred times higher than the best organic dyes, make them excellent sensing platform [6,16], which has been reviewed elsewhere [5,17]. For sensing and imaging in living cells, the AuNPs have been shown to confer greater stability of DNAs against degradation [18,19] and allow delivery of DNA into cells [20]. The first such demonstration is intracellular ATP detection in HeLa cells using DNA aptamer-functionalized AuNPs (Figure 2a) [21].…”

“…20 In these methods, the increased stability of the hybridization (or the affinity between the complementary oligonucleotides) has been achieved by substantially increasing the local concentrations of the oligonucleotides via assembling the two complementary oligonucleotides to a target molecule. Mirkin, et al has also reported the improved binding affinity for DNA-DNA interaction in gold nanoparticles attached DNAs (DNA-AuNPs) system, including sharp melting transitions [21][22][23][24][25] and increased binding constants. [26][27][28] The equilibrium binding constants (K eq ) of DNA-AuNPs with its complementary DNA are several orders of magnitude higher than their individual component DNA strands.…”
