A Disease Resistance Gene in Arabidopsis with Specificity for Two Different Pathogen Avirulence Genes

Abstract: The RPSS and RPMl disease resistance loci of Arabidopsis confer resistance to Pseudomonas syringae strains that carry the avirulence genes avrB and avrRpm7, respectively. We have previously shown that RPS3 and RPMl are closely llnked genetically. Here, we show that RPSS and RPMl are in fact the same gene. We screened a mutagenized Arabldopsls population wlth a R syringae strain carrying avrB and found 12 susceptible mutants. AI112 mutants were also susceptible to an lsogenic strain carrying avrRpml, indlcating… Show more

“…Wild-type plants expressed gene-for-gene resistance against PstDC3000(avrB) due to the presence of the CC-NBS-LRR gene RPM1 (28,29). Although the response of the atgsnor1-1 line was indistinguishable from wild-type plants, RPM1-mediated protection was abolished in the atgsnor1-3 mutant line (Fig.…”

“…In Col-0 plants, the product of the bacterial avirulence gene avrB is recognized by a single dominant R gene, RPM1 (28,29). Thus, PstDC3000 expressing avrB elicit a gene-for-gene resistance response in this Arabidopsis accession, leading to a reduction in bacterial growth.…”

“…Pseudomonas syringae pv. tomato (Pst) DC3000 expressing the avirulence gene, avrB (18), is recognized by the R protein RPM1 (18,19), triggering the expression of race-specific disease resistance in Arabidopsis accession, Col-0. Challenge of this plant line with avirulent PstDC3000(avrB) modified the S-nitrosylation status of a series of proteins including A. thaliana SA-binding protein 3 (AtSABP3; At3g01500) (20).…”

“…avrB was originally cloned from P. syringae pv. glycinea and later was found to mediate the elicitation of an RPM1-dependent HR and resistance in A. thaliana Col (44). Unlike DC3000, hrpA and hrpS mutants fail to elicit an HR in Rio-Grande PtoR (Table 1).…”
