A direct protein-protein interaction is involved in the suppression of β-catenin transcription by retinoid X receptor α in colorectal cancer cells

Abstract: This manuscript has been published online, prior to printing.Once the issue is complete and page numbers have been assigned, the citation will change accordingly.Using both mammalian two-hybrid assay in vivo and immunoprecipitation in vitro we found that retinoid X receptor α (RXRα) directly interacted with β-catenin and suppressed β-catenin transcriptional activity and protein expression in colorectal cancer cells. But, reduction of RXRα by small interfering RNA upregulated β-catenin transcriptional activity … Show more

“…Overall, Achilles tendons lacking Sparc are significantly thinner compared to their age-matched wild type counterparts and seem less densely packed based on histological evaluation. Further, they are characterized by smaller fibrils, reminiscent of what has been described for the dermis of Sparc−/− mice17. Our findings also confirm previously published results that fibril diameters in healthy-aged wild type tendons follow a bimodal distribution with an increase in larger diameter fibrils43.…”

“…2, GAAATCAAAGTCAG ATGCTA. As described previously [16], 1.5 × 10 5 cells were plated per 6‐well plate in media containing 10% FBS to reach 50% confluency, and transient transfection of the siRNA was performed to result in a final siRNA concentration of 100 nM for SW620 cell line. The cells were harvested for Western blot analysis.…”

“…As described previously [16], whole‐cell lysates were incubated with specific AEG‐1 antibody at 4°C overnight using Catch and Release reversible Immunoprecipitation system (Upstate Biotechnology, Lake Placid, NY), followed by immunoblotting probed with anti‐β‐catenin.…”

“…Both plasmids were purchased from Upstate Biotechnology (Lake Placid, NY, USA). As described previously, 20 LS174, SW480 and A549 cells were cotransfected with 2 µg of each plasmid. To correct for differences in transfection efficiency, cells were cotransfected with a Renilla reporter plasmid using LipofectAMINE 2000 (Invitrogen, Carlsbad, CA, USA) according to the manufacturer's instructions.…”
