A detailed triglyceride analysis ofLesquerella fendleri oil: Column chromatographic fractionation followed by supercritical fluid chromatography

Abstract: The triglyceride structure of oil from Lesquerella fendleri, a potential new U.S. crop, rich in C20 hydroxy fatty acids, was examined by silica gel column chromatographic fractionation followed by supercritical fluid chromatography. The analysis confirmed previous findings derived by our research group, but provided further detail. The analysis demonstrated the presence of trihydroxy triglyceride, which contained all of the oil's C78 hydroxy acyl groups (present at less than 0.5% in the oil). Lipolysis indicat… Show more

“…It may therefore not be surprising, that the structures of FA and the biosynthetic pathways may also be different. Five minor epoxy-and hydroxy-FA reported earlier [23] were not detected in this study as well as other studies [7][8][9][10]. Perhaps because the sample in this earlier study [23] was exposed to higher temperature than in other studies [7][8][9][10].…”

“…TAG and the contents (%) in Physaria fendleri oil were reported to have the highest content of TAG as OH20-18-OH20 (73.3 %, double bond not specified) by supercritical fluid chromatography [8]. The FA and their contents in Physaria fendleri oil reported [7][8][9] were similar. Atmospheric pressure chemical ionization mass spectrometry (APCI-MS) coupled to reversed-phase high-performance liquid chromatography (HPLC) was used for direct analysis of intact acylglycerols (AG) from the seed oils of Physaria fendleri, Physaria gordonii and castor [10].…”

“…Recently, enzymatic polymerizations have received much attention as a new methodology for polymer synthesis 8–14. The enzymatic polymerizations are performed in aqueous media or in bulk in the absence of heavy metal catalysts, which is a particular advantage 15.…”

“…The former method provided very good resolution of non-, mono-, di-and tri-hydroxylacyl (i.e., -ricinoleyl) TAG, and ricinoleic acid. Peak positions for the former three were confirmed using Lesquerella fendleri oil as a standard, due to its prominence of mono-and di-hydoxyacyl TAG [21]. Undecenoic acid appears as a shoulder at 5.0 min, but disappears during the time course of reaction due in part to its consumption, and to the incommensurately large decrease of its detector response with a small decrease of concentration (detector signal αconcentration 5.5 ).…”
