A decellularized flowable placental connective tissue matrix supports cellular functions of human tenocytes in vitro

Abstract: Purpose Injectable connective tissue matrices (CTMs) may promote tendon healing, given their minimally invasive properties, structural and biochemical extracellular matrix components, and capacity to fill irregular spaces. The purpose of this study is to evaluate the effects of placental CTMs on the cellular activities of human tenocytes. Decellularization, the removal of cells, cell fragments, and DNA from CTMs, has been shown to reduce the host’s inflammatory response. Therefore, the authors … Show more

“…Assessment of cell adhesion to AMs was performed in accordance with previous reports. 37 HCECs at passage 4 (P4) were cultured to 80% confluence in 10 cm cell culture dishes following the manufacturer's instructions. Cells were rinsed once with 5 ml phosphate‐buffered saline (PBS)/dish.…”

“…The prepared BF-ECM was characterized by the quantification of the residual DNA, SDS-PAGE, and immunofluorescent staining using antibodies against type I collagen or fibronectin. All characterization procedures followed the published methods and are described briefly below [22,39,40].…”

“…The adhesion and growth of cells (BUSCs expanded on BF-ECM or TCP) in the presence of different percentages of bovine fetal serum (FBS) were monitored as described [39]. Briefly, 2 × 10 4 cells were added per well (2.6 mg/cm 2 ) of a 48-well TCP.…”

“…Human placental-derived biomaterials have been increasingly recognized for their therapeutic applications across multiple diseases with consistent safety and efficacy profiles being observed [ 15 ]. For example, Interfyl®, a decellularized flowable placental connective tissue matrix product, is applied in wound healing [ 16 ], while BIOVANCE®, a decellularized and dehydrated human amniotic membrane allograft, supports tissue repair [ 17 ]. Serial studies have investigated different human placental tissue products in animal models of OA [ 18 23 ].…”
