A Critical Region in the FlaA Flagellin Facilitates Filament Formation of the Vibrio cholerae Flagellum

Abstract: is a Gram-negative bacterium with a monotrichous flagellum that causes the human disease cholera. Flagellum-mediated motility is an integral part of the bacterial life cycle inside the host and in the aquatic environment. The flagellar filament is composed of five flagellin subunits (FlaA, FlaB, FlaC, FlaD, and FlaE); however, only FlaA is necessary and sufficient for filament synthesis. is transcribed from a class III flagellar promoter, whereas the other four flagellins are transcribed from class IV promoter… Show more

“…However, only FlaA cells are deficient in both motility and flagellar polymorphism [53]. In common with S. putrefaciens , polar-flagellated bacteria possess multiple flagellins for flagellar polymorphism and migration in structured environments [54-56]. These data support our idea that the ability of bacteria to swim in structured environments is driven by flagellar polymorphism.…”

“…Genetic universal tool has been developed on suicide T vector and PBAD promoter system for rapid and efficient gene deletion in Vibrio species (20). Varied concentration of arabinose has been used as an inducer, which ranged from 0.1% to 0.2% for V. cholerae research (10,21). However, in case of expression of cloned protein, use of arabinose as high as 20% has also been reported (22).…”

“…FlaA is the only flagellin essential for the filament synthesis and the motility in V . cholerae , the four others being dispensable [19, 20]. In addition, V .…”
