A Cost-Effective Way to Produce Gram-Scale 18O-Labeled Aromatic Aldehydes

Abstract: Obtaining 18O-labeled organic substances is of great research importance and also an extremely challenging work. In this work, depending on the reversed Knoevenagel reaction, 18O-labeled aromatic aldehydes (3a–3x) are successfully obtained with high total yields (52–72%) and sufficient 18O abundance (90.90–96.09%).

“…Hence, deuteration is an important research issue in medicinal chemistry as well [10,11]. In addition, 2 H-, and even 14 C-and 18 Oisotope derivatives of furfural, especially of the aldehyde group, have been synthesized before [10,12]. As early as 1973, D.J.…”

“…Different conditions and solvents were reported in the literature for this reaction, however, many of them required long reaction time and/or expensive catalysts. Thus, various approaches for synthesis were reported: in methanol at room temperature for 12 h up to five days, [10] in refluxing ethanol for 4-12 h, [6,[11][12][13] in refluxing ethanol/piperidine/acetic acid for 4 h, [3] in refluxing ethanol/ piperidine for 5-10 h, [2] in aqueous NaOH solution for 12 h at room temperature, [5] in refluxing water for 2-12 h. [7,14,15] The reaction was reported in water using different catalysts such as L-tyrosine in 10 min. at room temperature, [1] and 2,2'-bipyridinium dihydrogen phosphate in 7-15 min at 70 °C.…”
