A core–BRAF35 complex containing histone deacetylase mediates repression of neuronal-specific genes

Abstract: BRAF35, a structural DNA-binding protein, initially was identified as a component of a large BRCA2-containing complex. Biochemical analysis revealed the presence of a smaller core-BRAF35 complex devoid of BRCA2. Here we report the isolation of a six-subunit core-BRAF35 complex with the capacity to deacetylate histones, termed the BRAF-histone deacetylase complex (BHC), from human cells. BHC contains polypeptides reminiscent of the chromatinremodeling complexes SWI͞SNF and NuRD (nucleosome remodeling and deacet… Show more

“…The LSD1/Aof2-Ring1B/Rnf2 complex(es) remain to be characterized fully, but preliminary evidence suggests it is a new entity not identified previously perhaps because it contains a relatively small fraction of Ring1B/Rnf2 (and most likely LSD1/ Aof2 too) in the cell. In fact, this histone demethylase has been identified in a variety of subsets of the Braf-HDAC (BHC) repressing complexes (55)(56)(57)(58), but our analysis of Ring1B/ Rnf2 complexes did not detect either Braf35 or BHC80, two of the components of core BHC complexes (55,58). Thus, our data suggest that the LSD1/Aof2-Ring1B/Rnf2 complex is a novel complex not related to BHC.…”

“…Coordinating the recruitment of chromatin-remodeling proteins by EMSY strengthens the link between BRCA2 and chromatin repair (17,32). This is not surprising given that DNA repair, like transcription, is a process that is challenged by chromatin structure.…”

“…A key RAS effector pathway involves the kinase cascade RAF/MEK/ERK (MEK: MAP/ERK kinase; ERK: extracellular signal related kinase). In the quest to understand how RAS transmits extracellular growth signals, the MAP kinase (MAPK) pathway has emerged as an important route between membrane-bound RAS and the nucleus Hakimi et al, 2002, Leder et al, 2002Gire and Wynford-Thomas, 2000).…”
