A Conserved E7-derived Cytotoxic T Lymphocyte Epitope Expressed on Human Papillomavirus 16-transformed HLA-A2+ Epithelial Cancers

Abstract: Human Papillomavirus 16 (HPV-16) has been identified as the causative agent of 50% of cervical cancers and many other HPV-associated tumors. The transforming potential/tumor maintenance capacity of this high risk HPV is mediated by two viral oncoproteins, E6 and E7, making them attractive targets for therapeutic vaccines. Of 21 E6 and E7 peptides computed to bind HLA-A*0201, 10 were confirmed through TAP-deficient T2 cell HLA stabilization assay. Those scoring positive were investigated to ascertain which were… Show more

“…By employing predictions as well as binding assays, and analyzing only non‐cysteine‐containing peptides, we reduced the number of possible HPV16 E6/E7‐derived HLA‐A2‐binding peptides to 17 actual binders, of which 9 had never been described before (Figure 2). The binding affinity results for already reported peptides were in accordance with our results, although they were tested with different in vitro assays 12, 13…”

“…Out of 14 tested donors, 8 showed reactivity against any of the tested peptides, indicating prior exposure to HPV16. Interestingly, the highest and most frequent responses were observed against E7 11–19 , which is the only peptide already detected to be presented on the cell surface of HPV16 + cells in a previous study 13. The overlapping peptide E7 12–19 also showed responses in four donors, albeit slightly weaker than the ones against E7 11–19 .…”
