A connection between MAPK pathways and circadian clocks

Abstract: Circadian clocks and mitogen-activated protein kinase (MAPK) signaling pathways are fundamental features of eukaryotic cells. Both pathways provide mechanisms for cells to respond to environmental stimuli, and links between them are known. We recently reported that the circadian clock in Neurospora crassa regulates daily rhythms in accumulation of phosphorylated, and thus active, OS-2 MAPK, a relative of mammalian p38 MAPK, when cells are grown in constant conditions. In the absence of acute stress, rhythmical… Show more

“…Moreover, the ERK1/2 MAPK pathway is also involved in regulating circadian rhythms in SCN and plays an important role in the clock-resetting mechanisms of mammalian circadian clock (40,44,45). Interestingly, mTOR activity also exhibits circadian variations in SCN (46) and has been reported to be crucial for regulating protein synthesis by modulating the phosphorylation of 4EBP and mRNAs with 5Ј-TOP motifs (47,48).…”

“…Based on clock control of the OS MAPK pathway (15,23) and OS-2-dependent phosphorylation of RCK-2 ( Fig. 1), we predicted that the circadian clock regulates the accumulation of P-RCK-2 in constant environmental conditions.…”

“…The list includes the nine components of the three MAPK cascades (24,42,49,53,56,68), PKA (5,32), PKC (23), checkpoint kinase 2 (73), ATM (Ataxia telangiectasia mutated; mus-21) (87), ATR (ATM and Rad3-related) homologs (mus-9) (87), the NDR (nuclear Dbf2-related) family of S/T kinases, cot-1 and dbf-2 (20,95), and others (see Table 1 for a complete list). Characterized Neurospora kinases have been shown to regulate a diversity of cellular functions, including polarized growth, hyphal fusion, asexual sporulation (conidiation), female sexual development, stress regulation, DNA damage responses, and the circadian clock (4,18,20,22,60,68,73,84,87,95).…”
