A comprehensive survey of the laminins and collagens type IV expressed in mouse Leydig cells and their regulation by LH/hCG

Abstract: Extracellular matrix (ECM) proteins have been shown to alter Leydig cell steroidogenesis in vitro, substantiating the hypothesis that Leydig cell steroidogenic activity and matrix environment are interdependent events. However, the nature of the ECM components synthesized by Leydig cells and their regulation by LH/human chorionic gonadotropin (hCG) remain unknown. Here, we examine the occurrence of the 11 laminin subunits and the 6 a chains of collagen IV (COL4A1-6) by RT-PCR in Leydig cells cultured with or w… Show more

“…1A, green and red arrowheads respectively), which is in agreement with previous studies. It is known that collagens including Col IV are expressed by Leydig cells in the mouse testis 33 and endothelial cells as well as basement membrane of microvessels 34 - 37 . Next, we investigated the upstream protease(s) that could possibly cleave the NC1 domain from Col IV in the basement membrane.…”

“…Briefly, first‐strand cDNAs were synthesized from 1 μg of total RNA in the presence of 100 U of Superscript II (Invitrogen, Eragny, France) and a mixture of random hexamers and oligo(dT) primers (Promega, Charbonnieres, France). Real‐time PCR assays were performed in duplicates for each sample with a Rotor‐Gene 6000 (Corbett Research, Mortlake, NSW, Australia), as described elsewhere (Mazaud Guittot et al. , 2008).…”

“…Melting curve analyses were performed immediately following the final PCR cycle to verify the specificity of the PCR product by checking its Tm. Rpl19 (ribosomal protein L19), and Gusb (glucuronidase beta) genes were chosen as references for normalizing target gene s in the testis (Mazaud Guittot et al. , 2008) and liver (unpublished data from the laboratory) respectively.…”

“…Such synthesis would be related to the control of steroidogenesis in which the α 1 and α 2 collagen chains appear to be structural in nature, while collagen α 3 α 4 α 5 and α 5 α 5 α 6 are regulated by gonadotropins in normal Leydig cells and therefore not in developing Leydig cells (Mazaud Guittot et al. ).…”
