A comprehensive pathway map of IL-18-mediated signalling

Abstract: Interleukin-18 (IL-18) is a member of the IL-1 family of cytokines and was initially described as an IFN-γ-inducing factor derived from anti-CD3-stimulated T-helper (Th)1 cells. IL-18 plays a significant role in the activation of hematopoietic cell types mediating both Th1 and Th2 responses and is the primary inducer of interferon-γ in these cells. The biological activity of IL-18 is mediated through its binding to the IL-18 receptor complex and activation of nuclear factor-κB (NF-κB), culminating in the produ… Show more

“…Still, IL-12 is a common feature of all Th1 differentiation protocols analyzed and was therefore not looked at in detail in this study. In contrast, the IFNγ-inducing factor IL-18 [ 50 ]—having been shown to play a role in both MS [ 58 ] and EAE [ 59 , 60 ] and to contribute to IFNγ-induction in Th1 cells [ 50 , 61 ]—was only used in some Th1 differentiation protocols. We therefore investigated the influence of IL-18 during the differentiation of Th1 cells on the eventual EAE course.…”

“…In addition, the interleukin 18 (IL-18) signaling pathway was triggered, which correlated with the activation of NF-κB, culminating in the production and release of several cytokines, chemokines and cellular adhesion molecules, as well as mitogen-activated protein kinases (MAPKs) activation. IL-18-mediated signaling acts as one of the vital components of the immunomodulatory cytokine networks involved in host defense, inflammation and tissue regeneration [80]. In addition, inflammatory bowel disease (IBD) is over-represented, probably due to DEGs associated with the innate immune response [81].…”

“…Generally poised as the activator of procaspase-1, inflammasomes are cytosolic multi-protein complexes that predominantly engage with innate immunity defense via activation of pro-interleukin (IL)-1β and pro-IL-18 cytokines ( 4 ). The active form of IL-18 is essential for interferon-gamma (IFN- γ ) production, which augments the cytotoxicity potential of natural killer (NK) cells and T cells ( 5 ). Active caspase-1, the pore-forming protein gasdermin D (GSDMD) that disrupts the osmotic potential and results in pyroptosis, is a form of programmed cellular death ( 6 ).…”
