A component of the chloroplastic protein import apparatus is targeted to the outer envelope membrane via a novel pathway.

Abstract: A chloroplastic outer envelope membrane protein of 75 kDa (OEP75) was identified previously as a component of the protein import machinery. Here we provide additional evidence that OEP75 is a component of protein import, present the isolation of a cDNA clone encoding this protein, briefly describe its developmental expression and tissue specificity, and characterize its insertion into the outer envelope membrane. 0EP75 was synthesized as a higher molecular weight precursor (prOEP75) which bound to isolated chl… Show more

“…For control assays, an equal number of chloroplasts were resuspended in resuspension buffer without ATP and the ATP regeneration system and incubated at 22°C in the dark. An equal amount of chloroplasts were taken at 0, 1.5, and 3 h and analyzed by SDS-PAGE and immunoblotting using antibodies against Toc75 (Tranel et al, 1995) and GS2 (Agrisera; AS08 296). The primary antibodies were used at 1:4,000 and 1:5,000 dilutions for Toc75 and GS2, respectively.…”
