A complete MAPK cascade, a calmodulin, and a protein phosphatase act downstream of CRK receptor kinases and regulate Arabidopsis innate immunity

Abstract: Mitogen-activated protein kinase (MAPK) cascades are critical signal transduction modules in stress responses, but how their composition and mode of activation induces a stress response is poorly understood. We showed in Arabidopsis that CRK21, a cysteine-rich receptor-like protein kinase (CRK), phosphorylates MAPK kinase kinase 20 (MKKK20) and thus directly activates a novel MAPK cascade, consisting of MKKK20, the MAPK kinase MKK3, and the MAPK MPK6. Furthermore, the protein phosphatase PP2C76 and the calmodu… Show more

“…MAPK3K13 and MAP3K14 may also be involved in a second MAPK cascade through a distinct MAP2K. For example, MAP3K20, which also belongs to subclade III of the MAP3K family together with MAP3K13 and MAP3K14, may act upstream of MKK5-MPK6 and MKK3 (Li et al, 2017;Sözen et al, 2020;Bai et al, 2022). To test these hypotheses, it would be interesting to characterize the MKK3-independent roles and molecular mechanisms by further comparing the phenotypes of plants impaired in MKK3 function and plants impaired in MAP3K13 and MAP3K14, as well as to identify the targets and interactors of MAP3K13 and MAP3K14, for example by performing phosphoproteomics and identifying interacting proteins.…”
