A Complete Genome Screen in Sib Pairs Affected by Gilles de la Tourette Syndrome

Abstract: Gilles de la Tourette syndrome is a neuropsychiatric disorder characterized by waxing and waning multiple motor and phonic tics with a complex mode of inheritance. Previous attempts, which used large multigenerational families to localize susceptibility loci, have been unsuccessful. In this report, the results of the first systematic genome scan, using 76 affected-sib-pair families with a total of 110 sib pairs, are summarized. While no results reached acceptable statistical significance, the multipoint maximu… Show more

“…Mathews et al ( 2007 ) gathered lifetime data for children and adults diagnosed with Tourette’s disorder from two genetically isolated samples using the Yale Self Report Form (YSRF; Tourette Syndrome Association International Consortium for Genetics 1999 ). In each sample, they found evidence for two symptom clusters, a simple tic cluster and complex tic cluster, and that the complex tic cluster was positively associated with tic severity, impairment, and comorbid obsessive compulsive disorder (OCD) symptoms.…”

“…We also reviewed the Diagnostic Confidence Index ( Robertson et al , 1999), the YGTSS ( Leckman et al , 1989; Storch et al , 2005), and the parent and child self-report forms used by the Tourette Syndrome Association International Consortium for Genetics (1999). An expert in psychiatric epidemiology (CWS) developed the questions that would be posed, in consultation with a movement-disorders-trained neuropsychiatrist (KJB).…”

“…Although large linkage studies have been performed, a genome-wide screen for linkage using 386 markers did not show a limit of detection (LOD) score of more than two, 41 nor did a genome -wide screen based on 110 sib pairs, show significant loci. 42 A sample from a French-Canadian family (127 members, 20 to 40 affected) showed a LOD score greater than three on Hq23. 43 …”
