A Competitive Microflora Increases the Resistance of Salmonella typhimurium to Inimical Processes: Evidence for a Suicide Response

Abstract: The presence of a viable competitive microflora at cell densities of 108 CFU ml−1 protects an underlying population of 105 CFU of Salmonella typhimuriumml−1 against freeze injury. The mechanism of enhanced resistance was initially postulated to be via an RpoS-mediated adaptive response. By using an spvRA::luxCDABEreporter we have shown that although the onset of RpoS-mediated gene expression was brought forward by the addition of a competitive microflora, the time taken for induction was measured in hours. Sin… Show more

“…In the field of food microbiology, specific recovery media and procedures that allow maximal recovery of sublethally injured cells have been developed. A factor that seems to be particularly important during recovery is oxidative stress (Aldsworth et al 1998;Dodd and Aldsworth 2002). Oxidative stress in bacterial cultures is caused by reactive oxygen species (ROS), which include hydrogen peroxide (H 2 O 2 ), the superoxide radical (O À 2 ), the hydroxyl radical (OH°) and singlet oxygen ( 1 O 2 ).…”

“…Standardized mid-exponential phase Salm. typhimurium LT2[pSB367] cultures were prepared as previously described (Aldsworth et al 1998) and inoculated into 50 ml volumes of each type of medium to an initial density of 10 5 cfu ml −1 . The cultures were then incubated aerobically at 37°C and 200 rev min −1 in an orbital incubator, and the bioluminescence and optical density at 600 nm were monitored at 30 min intervals using a Turner TD20e luminometer (Turner Designs Inc., Mount View, USA) and Gallenkamp Visi-Spec spectrophotometer, respectively.…”
