A comparison study of prabotulinumtoxinA vs onabotulinumtoxinA in myostatin‐deficient mice with muscle hypertrophy

Abstract: Botulinum toxin A (BoNT-A) is used clinically for various muscle disorders and acts by preventing the release of the neurotransmitter acetylcholine into the synapse space. Here, we compared the efficacy of prabotulinumtoxinA (PRA) and onabotulinumtoxinA (ONA) for the reduction in hypertrophy in myostatin-deficient (Mstn −/− ) mice. Two different BoNT-A products (2.5, 10 and 25 U/kg) were injected to paralyse the hindlimb for 2 months, after which sciatic nerve conduction study, 3D micro-CT, haematoxylin and eo… Show more

“…Generally, IncobotulinumtoxinA and OnabotulinumtoxinA may have close to a 1:1 dose equivalence ratio, but some dispute this 78 . PrabotulinumtoxinA and OnabotulinumtoxinA may have a 1:1 or higher ratio 71,72 . AbobotulinumtoxinA and OnabotulinumtoxinA seem to have a 2:1 or 2.5:1 equivalence ratio, although the variance of ratios used among studies ranges from 1.5:1 to 3:1 73‐77,79 .…”

“…65,66 Those that do attempt to compare brands have somewhat inconsistent and conflicting results, which may be due to a plethora of confounding variables that are difficult to control in addition to the lack of consensus regarding dose unit conversion ratios upon which they are based. 18,[65][66][67][68][69][70][71][72][73][74][75][76][77] Generally, IncobotulinumtoxinA and OnabotulinumtoxinA may have close to a 1:1 dose equivalence ratio, but some dispute this. 78 PrabotulinumtoxinA and OnabotulinumtoxinA may have a 1:1 or higher ratio.…”

“…Microscopic imaging and micro-CT imaging have also been employed to observe changes (such as injury or recovery) observed from the condition of mouse soleus muscle. Existing studies on imaging of this subject have been performed by identifying changes in the muscle shape due to hypertrophy in mice 13 , determining the effects of treatment on muscle atrophy 14 , and measuring the effects on sciatic neuralgia 15 using microscope or micro-CT.…”
