A COMPARISON OF THE METABOLISM IN VITRO OF [7α-3h] DEHYDROEPIANDROSTERONE SULPHATE AND [4-14c] PREGNENOLONE BY TISSUE FROM a HILUS CELL TUMOUR OF THE OVARY

Abstract: Tissue from a hilus cell tumour of an ovary removed from a 58-yr.-old virilized woman was incubated simultaneously with [7\g=a\-3H]dehydroepiandrosterone sulphate (DHA sulphate) and [4-14C]pregnenolone. The tissue was chopped and incubated in Krebs-Ringer bicarbonate glucose medium without the addition of cofactors. Both steroids were efficiently metabolized by the tissue; 78\m=.\5and 1\m=.\2 % of the pregnenolone was converted to androstenedione and testosterone respectively, whereas 34\m=.\6 % of the incubat… Show more

“…The prostate lobes were re¬ moved, weighed, minced with scissors, 300 fig of both testosterone and 5a-dihydrotestosterone (f 7/3-hydroxy-5«-androstan-3-one) added in 5 ml acetone and the mix¬ ture homogenized in a Silverson homogeniser. Steroids were extracted, purified by extensive thin chromatography and the specific radioactivities of the parent steroids and their derivatives determined (Fahmy et al 1968). …”

“…The tissue was blotted again and homogenised in buffer using a Griffiths ground-glass homogeniser. Homogenates were made up to 10 ml with distilled water, an aliquot taken for protein analysis (Lowry et al 1951) and acetone containing 300 «g various carrier non-radioactive steroids added to the remainder before determining the specific radioactivity of the steroids by techniques previously described (Fahmy et al 1968). …”

“…The following carrier steroids (200 ,ug of each) were then added: oestrone, 1 7f-oestradiol, oestriol, 16ao-hydroxytestosterone, 5c-dihydrotestosterone, 5ox-androstan-3ca,17/3-diol and 5oa-androstan-3,17-dione. Five vol of acetone were added and neutral, conjugate and phenolic fractions isolated by the method of Fahmy et al (1968).…”

“…Before extraction of metabolites, 500 ug of non-radioactive carrier of the steroids to be characterized were added to enable monitoring of procedural losses. Metabolites were then extracted as described by Fahmy et al (5) and separated into individual steroids by continuous elution thin layer chromatography (t.l.c.) for 2h on Silica gel HF 254 & 366 in chloroform:acetone (98:2).…”
