A comparison of manual and semi‐automated methods in the assessment of axonal injury

Abstract: Diffuse axonal injury (DAI) in the central nervous system is a common cause of post-traumatic coma and may result in varying degrees of disability up to and including the vegetative state. Experimental studies in man and animals have previously relied upon semi-quantitative grading systems for determining the relationship between the extent of DAI and the clinical features of patients. Using beta-amyloid precursor protein immunocytochemistry for the detection of DAI in sections of corpus callosum from 15 cases… Show more

“…4F). Cases H4, H7, and H8 had APP loads ranging from 0% to 0.5% in the posterior regions, corresponding to mild axonal damage or a rating of 1 on our previously established manual grading scheme (Gentleman et al, 1995(Gentleman et al, , 1999. Cases H1, H2, H3, and H5 had APP loads greater than 1% in at least one of the posterior regions examined.…”

“…In our previous study of the corpus callosum, the analysis of APP load was performed using a SeeScan image analyzer (Gentleman et al, 1999). Load measurements were defined by measuring the area occupied by APP immunostaining and dividing by the area of the sample field.…”

“…16 Briefly, tissue sections were digitized using a Nikon LS-2000 slide scanner. Regions of focal axonal damage were selected by density thresholding in Adobe Photoshop 4.0 and the areas were calculated and summated using the public domain NIH image program (developed at the U.S. National Institutes of Health and available on the Internet at http://rsb.info.nih.gov/nihimage/) (Figure 1; A to C).…”
