A Comparison of HLA Genotype with Inflammation in Uveal Melanoma

Abstract: The results before correction suggest that the HLA genotype may influence inflammation as indicated by HLA expression and macrophage infiltration in uveal melanoma. However, after correction this association did not prove significant.

“…This indicaties a proper function antigen presenting system, which one would not expect, as likely the most common method a tumor deploys for immune evasion is reducing their antigen expression by down regulation of HLA molecules [ 57 , 58 ] or altering the subtypes of HLA molecules expressed [ 59 ]. In earlier work we also showed that this latter is not the case [ 30 ]. Together, our data indicates that UM contain a proper functioning HLA antigen expression system, which should allow T-cell mediated tumor cell killing.…”

“…The current study population has previously been described [ 30 ], but only cases with material suitable for gene-analysis were included; each tumor sample was processed for conventional histopathological evaluation, including cell type assessment according to the modified Callender classification. The mean age at enucleation of the 28 patients was 62 years (median 68, range 28–84 years) and median follow up was 72 months (median 76, range 14–145 months).…”

“…This study on 235 Dutch UM patients and 2440 healthy controls did not find that HLA antigens contribute to an increased genetic predisposition to UM [60]. Additionally, van Essen et al determined whether specific HLA alleles are associated with increased inflammation: a decreased number of infiltrating macrophages was observed in HLA-A2 positive tumors, prior to correction for the number of antigens studied [61].…”

“…The origin of necrosis is unknown, but immunity may play a role in this process. [47][48][49] It is likely that necrosis reflects the presence of hypoxic areas with increased HIF-1a activity. Therefore, HIF-1a expression has been found to be a prognostic biomarker in patients with necrotic astrocytic and invasive bladder tumors.…”
