A comparison of chlordiazepoxide, bretazenil, L838,417 and zolpidem in a validated mouse Vogel conflict test

Abstract: We have established the VCT in C57BL/6J mice and validated this test behaviourally, physiologically and pharmacologically. The novel GABAA receptor ligand L838,417 was anxiolytic in this mouse model, and unlike the non-selective compounds, had no effect on unpunished drinking.

“…Our Vogel conflict method was originally validated with benzodiazepines in C57BL/6J mice, and we observed robust anxiolytic-like effects similar to those reported by Mathiasen and Mirza (2005); however, in the C57BL/6N mouse strain (parental background for the ␣ 2 ␦ mutants), benzodiazepine activity was blunted (http:// www.sfn.org/absarchive/abstract.aspx; Hain et al, 2001;Lotarski and Kinsora, 2004;Mathiasen and Mirza, 2005). It is well known that behavior and pharmacology in mice can be very sensitive to the strain used for testing (Crawley et al, 1997;Silva et al, 1997).…”

“…Significant motor effects were not observed [367]. L-838417 was also anxiolytic in murine Vogel conflict test [368]. Additionally, the compound exhibited anticonvulsant activity [369].…”

“…This test was a modification of that described by Vogel et al (1971) [26] and recently used by Mathiasen and Mirza (2005) [27]. The test cage (14.5×16×16 cm) was made from glass and had a metal grid floor.…”
