A Comparison of Automated Perfusion- and Manual Diffusion-Based Human Regulatory T Cell Expansion and Functionality Using a Soluble Activator Complex

Abstract: Absence or reduced frequency of human regulatory T cells (Tregs) can limit the control of inflammatory responses, autoimmunity, and the success of transplant engraftment. Clinical studies indicate that use of Tregs as immunotherapeutics would require billions of cells per dose. The Quantum® Cell Expansion System (Quantum system) is a hollow-fiber bioreactor that has previously been used to grow billions of functional T cells in a short timeframe, 8–9 d. Here we evaluated expansion of selected Tregs in the Quan… Show more

“…HF cartridges have been tested as cell proliferation chambers and demonstrated efficient expansion of adhesive cells, explained by a high inner surface area of the fibrils [ 32 ]. Interestingly, several studies also report the use of HFs for the expansion of suspension cells [ 33 , 34 ].…”

“…With full control of media flow rate, temperature and gas compositions afforded by the automatically fed cabinet, the Quantum system facilitates reduced manual operations thereby reducing the chance of contamination while improving reproducibility [73]. To date the platform has been used for, the expansion of CD3+ T-cell [75,76], MSCs [46,77], adipose stromal cells [78], neural stem cells [54], and iPSCs [79]. The DUET pump platform developed by FibreCellSystems is intended for laboratory scale HFBR systems.…”

“…Gas transfer happens passively in static systems, which limits oxygen availability in high volume vessels, as the diffusive flux of a gas is inversely proportional to the thickness of the liquid that needs to be permeated, according to Fick’s law and the McMurtrey model of oxygen diffusion [ 60 ]. Oxygen transfer may be limited in non-perfused bioreactors because low agitation rates are required to minimize shear stress on the lymphocytes and headspace aeration is also generally preferred for the same reasons.…”
