A comparative study of sertraline dosages, plasma concentrations, efficacy and adverse reactions in Chinese versus Caucasian patients

Abstract: This prospective 6-week study examined the differences in dosage and steady state plasma concentrations of sertraline in Chinese versus Caucasian depressed patients. Two groups of Chinese patients from different geographical sites and a group of Caucasian patients were evaluated with clinical measures during an initial dose of 50 mg/day, with subsequent doses adjusted clinically. The results of 17 Australian Chinese (ACHI), 13 Malaysian Chinese (MCHI) and 15 Australian Caucasians (AC) were analysed. Despite co… Show more

“…Future studies that also include men are warranted. Fifth, given the ethnicity-based differences in the pharmacodynamics and pharmacokinetics of psychotropic medications (38, 39), the current results from an ethnically homogenous study group need replication in a larger pool with different ethnic groups.…”

“…First of all, inter-ethnic differences in pharmacogenetics could affect antidepressant responsiveness. 28) Variations in gene allele frequencies can contribute to differences in antidepressant responses in different ethnic groups. 29) Over 90% of all drug metabolism could be accounted for by cytochrome P450 (CYP) enzymes, and 1A2, 2D6, 2C9, 2C19, and 3A4 account for 60% of metabolism.…”

“…Sertraline was commenced at 25 mg/day and increased by 25 mg/day every other day to achieve the optical dosage in ≀7 days, but the maximum dose would not exceed 100 mg/day. This sertraline dosing regimen would achieve a balance between the therapeutic benefits and adverse effect risk in the Chinese population …”
