A comparative study of microfocus CT and histomorphometry in the evaluation of bone augmentation in rat calvarium

Abstract: Microfocus computed tomography (micro-CT; R_mCT) is a dynamic noninvasive method for measuring bone regeneration. This study evaluated whether R_mCT was equivalent to histomorphometry in assessing bone augmentation. Two plastic caps of graft material with (experiment) or without hydroxyapatite (HA; control) were placed in the exposed calvaria of rats. Images of bone augmentation within the plastic caps were then taken using R_mCT. Histological sections were cut along the same plane as that used for the micro-C… Show more

“…The design of the skeletal envelope in the calvarium was according to the surgical procedure reported by us previously 1) . The advantages of our model include a lack of spontaneous regeneration and a standardized defect size by the use uniform plastic caps.…”

“…Although we have investigated the effect of various regenerative factors and biomaterials on GBA in rat calvarium, it was difficult to accurately measure the significant amount and height of newly generated bone in every instance [1][2][3] . Platelet-derived growth factor (PDGF) is a potent mitogen and chemotactic factor for mesenchymal cells.…”

“…In the AB group, a rat GBA model reported and standardized previously by our group was applied by setting plastic caps on the rat calvaria [12,13]. All operations were conducted under sterile conditions.…”

“…To overcome these limits, procedures using a bone substitute [12,13] or bioactive molecules [14,15] are used with the original GBR theory. Even if adequate BV is acquired successfully by the GBR procedure, a bony ridge in the desired shape is rarely obtained.…”
