A comparative study of histological and immunohistochemical methods for neurofibrillary tangles and senile plaques in Alzheimer's disease

Abstract: Several studies have demonstrated that the accurate visualization and quantification of pathological lesions in neurodegenerative disorders depend on the reliability of staining methods. In an attempt to gain a better assessment of the density and distribution of the neuropathological markers of Alzheimer's disease, we compared the staining efficiency of a modified thioflavine S protocol for neurofibrillary tangles (NFT) and senile plaques (SP) to different argentic impregnation techniques (Bielchowsky, Gallya… Show more

“…There were no statistically significant differences in the numbers of neurofibrillary tangles in several cortical regions among APOE3/3, -3/4, and 4/4 genotypes. The literature suggests that neurofibrillary tangles are endpoints of neuronal injury in a variety of neurodegenerative illnesses (37)(38)(39). The counts in the various cortical regions examined correlated strongly with duration of illness (P < 0.01) in the patients in this study.…”

“…Cells were immunostained with an antibody that recognizes total tau and counterstained with thioflavin-S. Thioflavin-S binding is a known indicator of insoluble protein aggregates with ␤-pleated sheet structure (38), and thioflavin-S readily stains the intracellular neurofibrillary tangles in Alzheimer's disease brain (39). Thioflavin-S-positive inclusions were not observed in cells transfected with either the fulllength or Asp-421-truncated tau constructs alone (Fig.…”

“…Silver staining of tissues was achieved using an adapted version of the Campbell-Switzer silver stain protocol (67). Briefly, tissue sections were treated with 2% ammonium hydroxide, rinsed in water, then placed in silver-pyridine-carbonate solution (0.5% silver nitrate, 0.4% potassium carbonate, 0.14% pyridine) for 40 min in the dark.…”
