A comparative study of glutathione and ascorbate metabolism during germination of Pinus pinea L. seeds

Abstract: The ascorbate and glutathione systems have been studied during the first stages of germination in orthodox seeds of the gymnosperm Pinus pinea L. (pine). The results indicate that remarkable changes in the content and redox balance of these metabolites occur in both the embryo and endosperm; even if with different patterns for the two redox pairs. Dry seeds are devoid of the ascorbate reduced form (ASC) and contain only dehydroascorbic acid (DHA). By contrast, glutathione is present both in the reduced (GSH) a… Show more

“…Similarly, in dry Pinus pinea seeds GSSG represented 50% of the total glutathione pool in embryos. This was reduced to less than 4% after 24 h of germination as a result of GSSG reduction by GR (Tommasi et al 2001).…”

“…Antioxidant enzyme activities were assayed at 4°C on nondenaturing polyacrylamide gels with a Mini-Protein II electrophoresis cell (Bio-Rad, Hercules, CA, USA) according to the following procedures (using the notation of Righetti 1983); CAT isozymes (7.5% T, 3% C; Anderson et al 1995), APX isozymes (10% T, 3% C; Lee and Lee 2000), SOD isozymes (12% T, 3% C; Beauchamp and Fridovich 1971), GR isozymes (7.5% T, 3% C; Anderson et al 1995), and DHAR isozymes (12% T, 5% C; Tommasi et al 2001).…”

“…Ascorbate and glutathione contents were determined according to Sgherri et al (2000) and Tommasi et al (2001), respectively. The content of hydrogen peroxide (H 2 O 2 ) was measured by monitoring the A 410 of titaniumperoxide complex following the method of Patterson et al (1984).…”
