A comparative study of bovine blood and milk neutrophil functions with luminol‐dependent chemiluminescence

Abstract: In this study, a technique was developed for the chemiluminescence (CL) measurement of bovine milk polymorphonuclear leukocytes (PMN). In the first study, the effects of cell number and the concentration of phorbol-12-myristate-13-acetate (PMA), luminol, latex bead particles, dimethyl sulphoxide (DMSO) and gelatin on the luminol-dependent cellular CL (LDCL) response were assessed with healthy cows in different stages of lactation. In the second study, the LDCL and in vitro bactericidal activity of blood and mi… Show more

“…The basal adhesion molecule levels of CD62L and CD18 at the surface of circulating PMN and the phagocytic activity of blood and milk PMN have been described to be similar during early lactation as compared to later stages of lactation [63,210]. Interestingly, ROS production and killing activity of blood and milk PMN were significantly decreased in early lactation [63,158,159,161]. Because PMN functions are markedly altered around parturition (Fig.…”

“…Bovine blood and milk PMN ROS production can be measured by chemiluminescence (CL; see [157][158][159]), which is associated with the bactericidal capacity of PMN [3,84,187,249]. Studies on CL kinetics in PMN revealed extra-and intracellular events during ROS production [25,49,69,73].…”

“…Stimulation of NADPH oxidase in the plasma membrane initiates hexose monophosphate activity. Bovine PMN release large amounts of superoxide (O 2 -) and hydrogen peroxide (H 2 O 2 ) when their plasma membrane is perturbed [46,100,109,183]. The O 2 -and H 2 O 2 formed on the cell surface and phagosomal membrane interact to form hydroxyl radical and singlet oxygen, key components of oxygen-dependent killing mechanisms of PMN.…”
