Intra and inter populational genetic variability in Maytenus ilicifolia Mart. ex Reiss. 1861, through RAPD markers

Maytenus ilicifolia is a medicinal plant largely used in the South Brazilian folk medicine. The aim of this study was to quantify the intra and inter populational genetic variability in three populations of M. ilicifolia, focusing on the genetic conservation of this species, which has been threatened by anthropic action. RAPD (Random Amplified Polymorphic DNA) markers were used to analyze 30 plants of each of the three populations collected in the Alto Uruguai Gaúcho region. Fourteen selected primers generated a total of 158 bands, 71.5% of which were polymorphic. The comparison of Jaccard's distances showed that the intra populational variation was higher than the inter populational variability, and cluster analysis allowed the separation of the three populations. Just 7.6% of the bands were specific of at least two populations. Data indicate that the analyzed M. ilicifolia populations represent a single genetic pool, and therefore any of the population thoroughly can represent the overall genetic variability of the species in the sampled region.Keywords: conservation, espinheira santa, native populations, molecular markers. Variabilidade genética intra e interpopulacional em Maytenus ilicifolia Mart. ex Reiss, utilizando marcadores RAPD ResumoMaytenus ilicifolia é uma planta medicinal bastante utilizada na medicina popular da região sul do Brasil. O objetivo deste estudo foi quantificar a variabilidade genética intra e interpopulacional em três populações de M. ilicifolia visando a conservação genética desta espécie, que se encontra ameaçada pela ação antrópica. Marcadores RAPD (Random Amplified Polymorphic DNA) foram utilizados para analisar 30 plantas de cada uma das três populações coletadas na região do Alto Uruguai Gaúcho. Foram selecionados 14 primers, que geraram 158 bandas, das quais 71,5% foram polimórficas. A comparação das distâncias de Jaccard mostraram que a variabilidade intra populacional foi maior que a interpopulacional, e a análise de agrupamentos permitiu a separação das três populações. Somente 7.6% das bandas foram específicas de pelo menos duas populações. Os resultados indicam que as populações de M. ilicifolia analisadas representam um único conjunto gênico, de tal forma que qualquer uma das populações pode representar a variabilidade genética geral da espécie na região.

RESUMO:Maytenus truncata Reiss (Celastraceae) é uma planta nativa da Bahia (Brasil), sendo conhecida como "espinheira-santa". É usada popularmente na forma de decoto das folhas (chás) como antiulcerogênico, similarmente à Maytenus ilicifolia, a verdadeira "espinheira-santa". O objetivo do presente estudo foi avaliar a atividade antiúlcera e cicatrizante, assim como o perfi l fi toquímico dos extratos brutos em acetato de etila e metanol da Maytenus truncata. A administração per os desses extratos nas doses de 120 mg/kg e 240 mg/kg reduziu a severidade da lesão gástrica induzida pelo estresse ao frio (-18 °C por 45 minutos) em ratos, com resultados mais signifi cativos para o extrato bruto obtido em metanol. A administração dos extratos provocou o aumento do pH. Os resultados obtidos na administração do extrato bruto em metanol não contrariam seu uso popular, não somente pela atividade observada, mas também por se tratar de um extrato de alta polaridade cujos princípios podem ser obtidos a partir de uma infusão, embora estudos clínicos devam ser realizados para confi rmação dessa hipótese. Per os administration of these extracts at 120 mg/kg and 240 mg/kg doses decreased the severity of gastric lesions induced by cold-restraint stress (-18 °C for 45 minutes) in rats, with more signifi cant results for the crude methanol extract. The administration of the extracts caused pH increase. A number of natural compounds have been used as immunomodulatory agents, enabling the function of the immune system to be modified by stimulating or suppressing it. There has been increasing interest in the study of therapeutic action of plant extracts regarding their immunomodulatory activity. The aim of this study was to identify and evaluate the action of extracts of the medicinal plants Calophyllum brasiliense, Ipomoea pes-caprae, Matayba elaeagnoides, Maytenus robusta, Rubus imperialis and Vernonia scorpioides on the development of spleen cells from mice, using the in vitro cellular proliferation assay. The cells, obtained by mechanical rupture of mice spleen (5x10 4 cells/mL), were incubated with methanol extracts (10, 50, 100 and 200 mg/mL) and phytohemagglutinin (PHA, 5 mg/mL). The basal control for proliferation consisted of cells alone, while the positive control consisted of cells and PHA. The cell culture was kept at 37 o C in 5% CO 2 for 72 hours, and cell proliferation was revealed by the blue tetrazolium reduction assay (MTT). The results were expressed as percentage of growth and were analyzed using the Kruskal-Wallis and Mann-Whitney tests. The C. brasiliense, I. pes-caprae and M. elaeagnoides extracts showed dose-dependent induction of cell proliferation, with a significant increase in cell proliferation (p<0.03) and percentage growth of 88.2%, 73.1% and 52.7%, respectively, suggesting T lymphocyte stimulation. By contrast, M. robusta, R. imperialis and V. scorpioides extracts showed significance only with a negative percentage of growth, suggesting inhibition of cell proliferation (p<0.04). Further biomonitoring studies will enable the fractions and isolated substances responsible for the immunomodulatory activities to be identified. Várias substâncias de origem natural têm sido utilizadas como agentes imunomoduladores, permitindo modificar a função do sistema imune e propiciando o estudo de atividades terapêuticas de extratos de plantas. Este trabalho objetivou identificar a atividade imunomodulatória dos extratos de seis plantas medicinais da flora brasileira, Calophyllum brasiliense, Ipomoea pes-caprae, Matayba elaeagnoides, Maytenus robusta, Rubus imperialis e Vernonia scorpioides, sobre a proliferação de células esplênicas de camundongos. As células esplênicas murinas obtidas por ruptura mecânica do baço (5x14 3 células/mL) foram incubadas com os extratos metanólicos das plantas (10, 50, 100, 200 mg/mL) e fito-hemaglutinina (PHA, 5 mg/mL). O controle basal de proliferação foi constituído de células apenas e o controle positivo formado por células e PHA. O cultivo celular foi mantido a 37 o C, 5% de CO 2 , 72 horas, com quantificação da proliferação celular pelo ensaio de redução do azul de tetrazólio.
