A comparative analysis of the distribution of glyprolines after their administration by different ways

Abstract: The distribution of the glyprolines Pro-Gly-Pro and Thr-Lys-Pro-Arg-Pro-Gly-Pro (Selanc) was analyzed and compared in tissues of rat organs after different ways of their administration using the peptides uniformly labeled with tritium. Comparative data on changes in concentrations of the peptides in the rat organs after their intraperitoneal, intranasal, intragastric, and intravenous administration are given. The intranasal administration of both peptides was shown to be optimal for the delivery of glyproline … Show more

“…The neurotropic effects of this dipeptide on stressed animals are no less pronounced than the effects of PGP [5]. GP is the main metabolite of Selanc, a synthetic peptide anxiolytic [1]. We also observed a depriming effect of Selanc in verticalization test, but this peptide did not modify 3 H-spiperone binding to D 2 receptors in vitro [7].…”

“…It enters unchanged into the circulation and crosses the blood-brain barrier [3]. However, just one thousandth of the injected peptide reaches the brain and is gradually hydrolyzed with the formation of GP as the main metabolite [1]. The neurotropic effects of this dipeptide on stressed animals are no less pronounced than the effects of PGP [5].…”

“…We studied the effects of Selank and GABA on changes in the mRNA levels of 84 genes involved in neurotransmission in the frontal cortex of rats 1 and 3 h after the intranasal administration of the compounds. The intranasal administration of Selank was shown to be optimal for delivery of peptide molecules in the CNS (Zolotarev et al, 2006 ; Ashmarin et al, 2008 ).…”

“…Selank dose of 300 μ g/kg was selected based on the data that this dose was the most effective therapy dose exerting anxiolytic action [ 10 , 13 ]. The intranasal administration of Selank was shown to be optimal for the delivery of peptide molecules in the CNS [ 14 , 15 ].…”
